A new anti-cancer method: "utilizing" cancer cell resistance

Due to the drug resistance produced by cancer cells, proteasome inhibitors have limited function in the treatment of cancer. Researchers at the Whitehead Institute have discovered a mechanism of resistance that occurs naturally in a variety of cancers that exposes the vulnerability of drugs and stimulates natural cell death.

The findings, published in the Proceedings of the National Academy of Sciences, are entitled to inhibit changes in the state of the 19 s proteasome unit-labeled cells in different cancers.

Hydrolases are large protein complexes that play an important role in regulating degradation and maintaining intracellular protein balance. When a cell begins to become cancerous, it creates tremendous stress and is responsible for maintaining the balance of the protein, a target called a proteasome inhibitor. Although proteasome inhibitors can effectively kill cancer cells in culture dishes (in vitro), they are greatly limited in drug resistance due to drug resistance.

The research team said: "At present, we have discovered an intuitive mechanism by which cells can obtain proteasome inhibitors from outside the body. In our report, the role of this mechanism in various human cancers is elaborated. In addition, we have determined that this mechanism is widely present in cells in different states and can be combined with existing drugs."

By analyzing thousands of cancer cells and tumors, the researchers found that cells that are resistant to proteolytic enzyme inhibitors are inhibited.

The report also pointed out that in addition to resistance to proteolytic enzyme inhibitors, the inhibition of proteasome subunit expression reflects a wide range of cellular genetic changes. This facilitates treatment with biomarkers.

The head of the research team said: "The resistance of cancer cells stems from a variety of mechanisms, genetic or epigenetic. With these findings, we can develop new strategies and new compounds as arrays, using them to be more prone to drug resistance. Used to effectively treat a variety of cancer types."
