Professor Zhang Qiwei, a co-author of Tsinghua University and the University of Texas at Dallas, said that the traditional FISH method is limited by various factors (including labeling ability and optical resolution) and it is difficult to obtain clear microscopic images of specific short fragments in the genome.

“Although this technique is promising in the study of the fine organization of chromatin domains, we hope to develop a new method: not only to further improve the resolution of the genome sequence, but also to avoid the cumbersome probe preparation process to the actual The limitations of the application, Professor Zhang Qiwei said, this new method can accurately target gene enhancers and promoters, interact, visualize in situ, and can be used to detect specific DNA in single cells in cancer and other diseases. One of the authors, Dr. Cao Bo from Shenzhen University, introduced the new method of molecular beacon (MB) probes (MB-FISH) on the self-built 3D-STORM platform. According to reports, the team is currently building and improving multi-color 3D-STORM, which will release greater potential in studying the 3D genomic structure of human normal and disease (eg cancer) cells.
